
A massive new genetic study is challenging the way we define mental illness, and confirming that the brain is where the real story begins. The Nature study, published in December 2025, analyzed genetic data from more than six million people, including over one million individuals diagnosed with at least one psychiatric disorder. Researchers examined fourteen conditions, ranging from depression, anxiety, and PTSD to ADHD, autism, bipolar disorder, schizophrenia, OCD, and substance use disorders.
Instead of finding completely separate genetic pathways for each diagnosis, the team discovered that many of these disorders share common genetic risk factors. These shared risks clustered into broader biological groupings tied to brain development and synaptic function (how brain cells communicate with one another).
In other words, the same genetic variants can increase vulnerability to multiple mental health conditions. The study also found that certain traits, such as suicidality and loneliness, were genetically linked across several disorder clusters.
The findings suggest that psychiatric conditions may be better understood as overlapping brain-based systems rather than isolated categories, which counters the way mental illness has traditionally been classified.

The DSM-5, psychiatry’s diagnostic manual, groups conditions by clusters of symptoms. But symptoms don’t always tell the full story.
This new research identified genetic clusters that do not neatly align with traditional diagnostic labels. That means conditions like depression, bipolar disorder, and schizophrenia may share overlapping biological systems that begin affecting the brain very early in life.
In clinical practice, this mismatch happens frequently, according to Dr. Daniel Amen, founder of Amen Clinics, who was recently interviewed by Fox News regarding the new genetic study.
“We’ve had patients come in with the same diagnosis—say, depression—but when we scan their brains, we see wildly different patterns. One may have overactivity in the limbic system, another underactivity in the prefrontal cortex, and another damage from trauma or toxic exposure. It’s possible that mental health care could shift from being reactive to proactive and preventive. “Imagine screening a 7-year-old for risk, optimizing their brain health early, and preventing future suffering,” Dr. Amen added.
And perhaps most importantly, stigma would decrease. That’s what happens naturally when mental health struggles are clearly understood as brain-based conditions, not character flaws. “If it’s your brain—not your fault—then there’s no shame, only opportunity for healing,” Dr. Amen explained.
